Course description

Bioavailability and biorelevant dissolution

The lecturers use their rich experience gained when solving real problems in the pharmaceutical R&D. In the introduction bioavailability of drugs and its optimization, i.e. modification of physico-chemical properties of a drug molecule in the chemical and formulation context, are considered. Advantages, disadvantages and application of individual pharmacopoeial dissolution methods that are used primarily in quality control are discussed in details.

The key part of the course is the problematics of so-called dynamic biorelevant dissolution, i.e. methods that provide results of formulation evaluation comparable with in-vivo studies. Simulation using dynamic biorelevant dissolution improves dissolution evaluation of oral drug formulations and increases the prediction value of dissolution. Using this approach, the number of unsuccessful bioequivalence studies can be minimized, and an appropriate formulation can be proposed. The review of existing dissolution systems and information how to stimulate physiological conditions of human gastro-intestinal tract for particular types of oral formulations to obtain the most objective results are provided. Also influence of food and interactions between drugs and excipients are mentioned. Examples of interactions between drugs administered in fixed combinations that influence bioequivalence are given.

The leader of the course is Dr. Dionigio Franchi, the former Director and Site Lead of the Pharmaceutical Development Department in the R&D of the original pharmaceutical company GlaxoSmithKline, Verona. His professional experience is the guarantee of the course quality. His lectures are in English, therefore course participants should understand English. Other lecturers are Natalia CsókováMartin Čulen and Josef Jampilek.
